Proofreaders

Proofreader are people who read over the script to see if there are any errors in grammar, punctuation, spelling, or placement/description of text (where which sentences go in which bubbles, thoughts, etc.). They make sure that every sentence makes sense before handing it to the typesetter.

Proofreaders have to be fluent in English and spot out all errors (things like 'she went to the the shop'). They also have to spot out any missing translations. So for this, it would be good if you had knowledge of the language of the original raws (ie Chinese, Japanese or Korean), although it is not compulsory. Knowledge of the story and characters of the manga would also be preferred although not compulsory.
